A search is now underway for a man missing after a factory explosion in South Australia, after a bomb disposal team secured the site.

Police can now confirm that two men were killed when an explosion ripped through the Quin munitions factory near Gladstone, 180 kilometres north of Adelaide.

Two other men received shrapnel wounds and have been hospitalised.

State Premier Mike Rann, says it's a terrible tragedy amidst the euphoria of the rescue of the Beaconsfield miners.

"On a day when of course there's been celebrations in Tasmania following the rescue of two miners there, here in South Australia there is a very close-knit community that has been hit terribly hard by this tragedy and they're going to need all the support they can get."

The factory is the site of a former army munitions depot, and makes explosives for the mining and defence industries.
