The 19-year-old was the Magpies' first pick at No.13 last November in the national draft.
Coach Nathan Buckley confirmed the selection before Wednesday's training and Quaynor will be their first debutant this season.
"I'm excited to get an opportunity and play my first game for Collingwood. It came as a bit of a surprise but I'm looking forward to getting out onto the MCG and playing my role," the speedy defender told the club website.
"Funnily enough, my mate had a dream about 10 days ago that I would debut against Hawthorn. It's pretty crazy how it's all happened."
Quaynor has played 10 VFL games this season amid speculation about when he would make his AFL debut.
The second-placed Magpies badly need a win after last Saturday night's horror loss to North Melbourne.
