The incident happened around 9am on Tuesday. Reports say thick back smoke which is about 30 meters high has filled the air.
Victoria's Police Minister Lisa Neville has confirmed that there were five people on board the plane when it crashed.
The Tullamarine and Calder freeways have been closed following the incident.
