Landa won two stages as he finished third in last year's race while with Astana and goes into the season's first Grand Tour in good form after winning the Giro del Trentino.
"The Giro d'Italia is my first big goal of the season and I'm proud to go there as leader of Team Sky. I feel like I'm coming into really good form and I'm confident I can perform well," Landa said.
"As usual the Giro will be a really tough race but we have a strong team and we're ready for it"
The 99th edition of the Giro begins on Friday 6 May with a 9.8km individual time trial around Apeldoorn in the Netherlands.
