Kangaroos flyer Valentine Holmes is in doubt for the start of Cronulla's NRL title defence after suffering a hamstring injury in the Sharks' 30-12 trial loss to Brisbane at Redcliffe on Saturday night.

Holmes joined a worrying Cronulla injury list that also included captain Paul Gallen, who was a last minute withdrawal with a neck concern.

Cronulla coach Shane Flanagan admitted the initial prognosis for Holmes was "not good" but scans were booked for the fullback on Sunday.

Holmes was assisted off the field after feeling a "slight strain" in his right hamstring while attempting to score in the 50th minute.

The Sharks will be sweating on the scan results for Holmes who was primed to step into the fullback role vacated by Ben Barba for their season opener against Brisbane on March 2.

Cronulla also have the World Club Challenge against Wigan in the UK on February 19.

"He just felt his hamstring grab a little bit when he jumped to (try and) score that try," Flanagan told the Sharks website.

"When he got back into the sheds, it wasn't as bad as first thought but it's not good.

"He'll have scans tomorrow morning so we will know exactly what's wrong."

On a tough night for the premiers, centre Jordan Drew suffered concussion in the five tries to two loss at Dolphin Oval.
