Slovenian Kevin Kampl put the hosts ahead with a left-footed curled effort from outside the box in the 16th minute before striker Timo Werner nodded in at the far post to complete a fine move 12 minutes later.
Wolfsburg upped the pressure towards the end of the first half but Leipzig keeper Peter Gulacsi twice came to the rescue to deny the visitors from close range.
His Wolves counterpart Pavao Pervan let a Werner shot slip through a minute after the restart but the forward was ruled offside.
Leipzig, missing injured top striker Yussuf Poulsen, continued to boss the midfield and launch quick attacks with Pervan making a world-class, one-handed save of a Werner shot in the 70th.
Werner also hit the woodwork late in the game, with the final scoreline being kind to the visitors.
Victory kept Leipzig firmly on course for a Champions League spot next season, in third place on 58 points with five matches left to play. Eintracht, on 52, host Augsburg on Sunday.
The top four finishers qualify for the lucrative Champions League group stage.
Second-placed Borussia Dortmund, who were playing later on Saturday against Mainz 05, are on 63.
Champions Bayern Munich, on 64, travel to Fortuna Duesseldorf on Sunday.
