Nanai-Williams has only featured at fullback since his debut for Samoa two years ago and for his Counties Manukau side in New Zealand, but he has been paired with Pele Cowley as the scrumhalf makes his first appearance for the national side since June last year.
They take the place of injured duo Kahn Fotuali'i and Auvasa Faleali'i who have been ruled out of the November tour, which also includes tests against Romania and England.
Loosehead prop Donald Brighouse will start his first test at tighthead with North Harbour lock Josh Tyrell to debut beside captain Chris Vui, who is among seven British-based players in the starting line-up.
