South African-born Du Preez will win a fourth cap at number eight in place of the injured Ryan Wilson who has not recovered from a shoulder injury.
Fagerson and Du Preez moved up from the bench last week and Simon Berghan and uncapped Luke Hamilton will take their places among the replacements at Murrayfield on Saturday.
