The Raptors overcame a 22-point deficit to beat the 76ers, who were again without centre Joel Embiid due to back tightness.
Simmons added six rebounds and four assists but had a game-high seven turnovers as the 76ers slumped to their fourth-straight game as their win-loss record now stands at 14-17.
As well as his points haul, DeRozan also set a career high with six six-pointers as the Raptors improved to 22-8, the second-best record in the eastern conference.
Kyle Lowry added 23 points to help the Raptors win their fifth straight while Robert Covington added 19 points and Dario Saric had 18 points, 10 rebounds and nine assists for hosts Philadelphia.
