A section of Melbourne's CityLink remains closed after a suspicious fire at a disused building near the historic Nylex clock site.

The Punt Road CityLink entry ramp near Gough Street in Cremorne was still closed on Sunday night following a blaze near the historic Nylex sign earlier that day.

Firefighters were called to the suspicious blaze in Cremorne just after midnight and crews arrived in less than six minutes to find the fire quickly spreading through the two disused buildings.

MFB declared the fire under control at 2.05am.

About 60 firefighters, 10 pumpers and three aerial units brought the fire under control.

Victoria Police say the exact cause of the fire is unknown and it is being investigated as suspicious.

Authorities expect road closures will remain at the Punt Road on-ramp to Citylink while works to repair or clear fire damage are undertaken.

VicRoads has advised commuters to take a detour Olympic Boulevard and Batman Avenue.

"It is not known when the entry ramp will reopen," according to the VicTraffic live feed.

The site's owners and developers, Caydon, says it has installed security measures to prevent trespassing and will be working with authorities in their investigations.

"We regret any inconvenience the fire may cause residents and commuter traffic, and are working with all authorities to make safe the site and its surrounds."
