A Sydney man was beaten and detained over a 12-hour period before he was killed over a dispute with his ex and her new lover, detectives will allege.

A child was also present while the man was held and driven around in a car, they will say.

Brett Watson, 41, suffered significant and brutal injuries to his body.

Raquel Hutchison, 36, and Paul Wilkinson, 35, each appeared in Penrith Local Court on Friday, charged with his October murder.

Neither applied for bail.

Police will allege Mr Watson's former partner Hutchison and her boyfriend Wilkinson ambushed him at his St Marys home in Sydney's west on October 20.

Over the next 12 hours he was allegedly held against his will and taken to various properties in St Marys, Mt Druitt and Minto before his body was dumped.

A third man, Daniel Greentree, an alleged associate of the couple, has been charged with being an accessory after the fact to murder.

Greentree, 42, who is on bail, appeared briefly in court on Friday.

A fourth person is wanted over Mr Watson's death.

Michelle Arthur, a friend of Watson and Hutchison, said outside the court she was shocked by the death.

Wilkinson's and Greentree's matters will return to court on April 17 while Hutchison will return to court on another matter on February 24.

Wilkinson said little as he sat in court while Hutchinson briefly clutched a set of blue rosary beads around her neck.

Police have released an image of a 1999-model white Ford sedan that was allegedly used to drive Mr Watson to Minto on October 20 in the hope people may recognise it.
