A 16-year-old boy has been charged with murder following the fatal stabbing of a man during a fight in Port Macquarie.

Police allege that during the fight about 1.40am on Sunday, a 16-year-old boy produced a knife and stabbed a 25-year-old man in the chest several times.

The boy and his friend then fled the scene in a ute while several bystanders went to the injured man's aid and attempted to revive him.

He was taken to hospital in a critical condition but died two hours later.

An 18-year-old man attended Kempsey Police Station on Sunday afternoon and was charged with being an accessory after the fact to murder. He will appear in court on Monday.

Several hours later, the 16-year-old also attended Kempsey Police Station where he was arrested and charged with murder. He was also refused bail to appear in a children's court on Monday.

Detectives also conducted search warrants at two premises in Dondingalong and South Kempsey where they seized a red Ford ute and other items for forensic examination.

Investigations are continuing.
