Two people have been killed in a Sydney house fire overnight.

Firefighters were called to the blaze in Penshurst at about 3.30am on Monday to find the house well alight.

There were grave fears for two people unaccounted for in the fire and police later discovered the bodies inside the house.

A post-mortem examination will be conducted to identify the remains.

The cause of the fire is not yet known.

Police later said two men managed to escape the weatherboard house and are being treated at St George Hospital for shock and smoke inhalation.

A couple who lived in the back of the house are missing.
